Fluc mRNA

1) Product description

Fluc-mRNA transfection into cells results in intracellular expression of firefly luciferase (luciferase) protein, which was originally isolated from the firefly Photinus pyralis. Firefly luciferase is a commonly used bioluminescence reporter molecule, which is commonly used in gene regulation, cell viability assays, ATP level analysis and in vivo imaging studies. Luciferase can catalyse the oxidation of the substrate luciferin to oxyluciferin in the presence of ATP and oxygen, and during the oxidation of luciferin, bioluminescence is emitted. The bioluminescence released during the oxidation of luciferin can then be measured at 560 nm by a chemiluminescence meter (luminometer) or a liquid flash detector. The bioluminescent system, luciferin and luciferase, allows extremely sensitive and efficient detection of gene expression.

The product is already a 5‘ end-capped, 3’ end poly(A) tailed and N1-Me-Pseudo UTP modified mRNA that mimics processed mature mRNAs in eukaryotes. mRNAs are capped in a co-transcriptional manner using 3-O-Me-GAG, which creates a Cap1 structure, which increases the stability and translation efficiency of the mRNA. The addition of the modified nucleotide N1-Me-Pseudo UTP reduces innate immune stimulation and increases mRNA stability. the addition of Poly(A) tails makes the mRNA more stable and improves the translation initiation efficiency of the mRNA.
